What is Zelboraf Used for?
Zelboraf (Vemurafenib) is a prescription medication used to treat adults with a serious type of skin cancer called melanoma that has spread or cannot be removed by surgery. It's specifically used when the melanoma has a BRAF V600 mutation, a genetic change that enables the cancer to grow. A lab test must confirm the presence of this mutation before starting treatment. Zelboraf is also used to treat Erdheim-Chester Disease (a rare blood cancer). The active ingredient in Zelboraf is Vemurafenib. Vemurafenib belongs to a class of medications known as kinase inhibitors (specifically targets BRAF V600E mutations). In healthy cells, the BRAF protein is a component of a signaling pathway that regulates cell growth and division. However, when the BRAF V600 gene is mutated, it remains constantly switched on, causing cells to grow uncontrollably. Vemurafenib works by blocking the mutated BRAF protein, helping to slow down or stop the growth of cancer cells with this mutation.

It is crucial to continue using this medication exactly as your doctor prescribes. Zelboraf comes as a tablet. Swallow the tablets whole with a full glass of water. Do not crush, chew, or break it. Take the medicine every 12 hours, at the same time each day. If you vomit shortly after taking Zelboraf, do not take another dose. Wait until your next scheduled dose time to retake the medicine. Zelboraf can increase your sensitivity to the sun. Wear protective clothing and use broad-spectrum sunscreen when outdoors. While using Zelboraf, you may need to undergo frequent blood tests to monitor your liver and kidney function. Your doctor will also order regular tests to monitor your skin condition while you are using the medicine and for several months after your last dose. You may also need to undergo frequent eye exams and an electrocardiogram (ECG or EKG). Vemurafenib can pass into body fluids, like urine, feces, and vomit. Caregivers should wear rubber gloves while cleaning up a patient's body fluids, handling contaminated trash or laundry, or changing diapers. They should wash their hands before and after removing gloves. Wash soiled clothing and linens separately from other laundry.

Women who can get pregnant and men with female partners who can get pregnant should use effective birth control to prevent pregnancy while using the medicine and for at least 2 weeks after the last dose. Inform your doctor if you are breastfeeding or planning to breastfeed. It is unknown if Vemurafenib passes into breast milk. However, there is an increased risk of harming a nursing infant. Do not breastfeed while using this medicine and for at least 2 weeks after your last dose. The safety and effectiveness of Zelboraf in children have not been established.
Tell your doctor about all the medications you use, including prescriptions, over-the-counter medications, herbal products, and vitamin supplements, to prevent drug interactions that might be fatal to your health. This drug interaction may make your drug less effective, cause adverse effects, or potentiate other drugs' actions. There may be a drug interaction between Zelboraf and other medications, such as ketoconazole, clarithromycin, rifampicin, carbamazepine, warfarin, and phenytoin.
